A previous study explored whether their chemicals could enhance the reprogramming process of human foreskin fibroblasts (hFFs) into hiPSCs. It was found that only RSC133 could significantly and dose-dependently increase the reprogramming efficiency of hFFs up to 10 μM when compared to the untreated control. Moreover, with an increase of RSC133 concentration to 20 μM, there was no change in cell viability, but it did not further enhance reprogramming efficienc. In addition, the continuous RSC133 treatment was the most effective way to increase reprogramming efficiency (about 3- to 4-fold). In combination with hypoxic conditions, RSC133 was able to synergistically increase the reprogramming efficiency by around 5- to 6-fold when compared to the untreated control.
